Rapamune is a brand-name drug that, in combination with cyclosporine and steroids, suppresses the immune system to prevent organ rejection after a kidney transplant. When tissue from one person is transplanted into another person, the body responds to it as a foreign object and mounts an immune response to it. Organ “rejection” happens when the immune system begins to damage the newly transplanted organ. Rapamune can be used in people who have had a kidney transplant, but should not be used in people who have had a liver or lung transplant.
The active ingredient in Rapamune, sirolimus, weakens the immune system so that it doesn't reject the newly transplanted organ. Sirolimus partially shuts down the immune system by blocking chemicals in the body that activate white blood cells, the body's first line of defense against foreign tissues. Your doctor will determine how long you need to take Rapamune.
Rapamune is first administered right after transplant surgery. Rapamune is taken along with cyclosporine and a steroid, at first, but the regimen may vary after several months, depending on risk. Rapamune is also used to treat a lung disease called lymphangioleiomyomatosis. Before taking Rapamune, talk to your doctor about your medical conditions and history, and all of the drugs you take, including prescription and OTC drugs, and vitamins or supplements.
Rapamune suppresses the immune system, the first line of defense against infections. So Rapamune lowers the body's ability to prevent or fight infections, putting patients at a higher risk of infections, including severe and life-threatening infections. These infections can include a viral infection that may cause kidney transplant failure or a brain infection that can cause disability or death. Immune suppression also increases the risk of certain types of cancer, such as skin cancer or lymphoma.
Other side effects include diarrhea, headache, nausea, vomiting, stomach pain, muscle or joint pain, mouth sores, constipation, pimples, swelling, and nosebleeds. Allergic reactions are rare. If you have symptoms of hives, difficulty breathing, or swelling of the face, lips, tongue, or throat, get emergency medical attention right away. Tell your doctor immediately if you have flu symptoms or a fever, burning upon urination, skin lesions, mental status changes, vision problems, weakness on one side of the body, pain around the transplant site, or problems with speech or walking.
