Prempro is a prescription drug that contains both estrogen and progestin as its active ingredients. Estrogen (conjugated estrogens in Prempro) and progestin (medroxyprogesterone acetate in Prempro) are female sex hormones used to treat symptoms of menopause. This medicine replaces estrogen that is no longer being made by the body. Estrogen helps with symptoms such as hot flashes and vaginal dryness, as well as prevents osteoporosis. Progestin is added to estrogen in hormone replacement therapy to lower the risk of uterine cancer in women with a uterus. Note: If a woman has had her uterus removed, there would be no need for her to use progestin.
Prempro is an oral tablet that is available in various strengths, and is taken once daily.
Common side effects of Prempro include abdominal or back pain, weakness, nausea, headache, breast pain/breast changes, vaginal bleeding/spotting, and depression.

Estrogen plus progestin:
Estrogen plus progestion treatment should not be used to prevent cardiovascular disease or dementia.
The Women’s Health Initiative (WHI) estrogen plus progestin substudy found increased risks of stroke, blood clots, heart attack, and breast cancer. The WHI Memory Study found estrogen and progestin to increase the risk of dementia in women 65 years and older.
Estrogen alone:
Women with a uterus who take estrogen (without a progestin) have a higher risk of endometrial cancer. (This is why women with a uterus are prescribed a progestin along with estrogen.)
Estrogen-alone treatment should not be used to prevent cardiovascular disease or dementia.
Estrogen alone can increase the risk of strokes and blood clots, as well as increase the risk of dementia in women 65 years and older.
Before taking Prempro, tell your doctor about all of your medical conditions. Prempro is not appropriate for everyone. Individuals who should not take Prempro include:
Women with undiagnosed genital bleeding
Women with breast cancer, suspected breast cancer, or history or breast cancer
Women with known or suspected estrogen-dependent cancer
Women with deep vein thrombosis, pulmonary embolism, or a history of either of these conditions
Women with certain medical conditions like stroke or heart attack or history of stroke or heart attack
Women with liver problems
Women who are pregnant or may be pregnant
Women with protein C, protein S, or antithrombin deficiency or other similar disorders
