What is Afinitor?
Afinitor is a brand-name prescription drug used to treat a variety of cancers, including breast cancer, neuroendocrine tumors, kidney cancer, and tuberous sclerosis.
Afinitor is used in combination with other drugs to treat advanced breast cancer in post-menopausal women. Advanced breast cancer is one that is aggressively growing and likely to spread to the rest of the body.
Neuroendocrine tumors (NET) are cancers that start in hormone-producing cells or nerves in the intestines, stomach, lung, pancreas, and other parts of the body. Afinitor is used to treat inoperable neuroendocrine tumors of the lung, intestines, or pancreas that are advanced (at risk of spreading to the rest of the body) or metastatic (have spread to the rest of the body).
Afinitor is used to treat advanced renal cell carcinoma. Renal cell carcinoma is a cancer of the tiny tubes in the kidney which then spreads to other kidney tissues.
Finally, Afinitor is approved for the treatment of a number of tuberous sclerosis complex (TSC) tumors. Tuberous sclerosis is a rare disorder that causes usually non-cancerous tumors (tubers) in several places around the body: skin, eyes, brain, kidneys, heart, lungs, and so on. TSC is the leading cause of genetic epilepsy; about 85% to 90% of patients with TSC have seizures. Only about 1 in 3 TSC patients respond at all to anti-seizure medications. Afinitor is approved to reduce TSC tumors in the kidneys (renal angiomyolipoma), TSC of the brain (subependymal giant cell astrocytoma, or SEGA), and to help reduce the number of partial seizures in TSC patients.
The active ingredient in Afinitor, abemaciclib, belongs to a class of anti-cancer drugs called kinase inhibitors. Abemaciclib works by blocking proteins on the surface of the cancer cells that, among other things, signal the cancer cell to divide and produce more cells. It's believed that these proteins, kinase receptors, become overactive in cancer cells. Blocking this protein slows or stops the growth of tumors.
Afinitor is administered as a tablet or oral solution (Afinitor Disperz) once daily until sufficient progress has been made against the cancer or the patient can no longer tolerate Afinitor.
What are the side effects of Afinitor?
Common side effects of Afinitor include diarrhea, nausea, vomiting, constipation, headache, loss of appetite, weight loss, dry skin, pimples, nosebleeds, mouth sores, temporary hair loss, arm pain, leg and joint pain, altered taste, and swelling. Afinitor may cause mental and mood changes, but these are rare.
The most serious side effects of Afinitor include high blood sugar, high cholesterol, high triglycerides, liver problems, kidney problems, kidney failure, lung problems (pneumonitis), and low white blood cell counts, putting the patient at risk for potentially severe infections.
Allergic reactions are rare, but could be severe, and include anaphylaxis, a dangerous drop in blood pressure. Watch for signs of an allergic reaction, such as swelling, rash, or trouble breathing, and seek medical help if you suspect you're having an allergic reaction to Afinitor.
