What is Tetrabenazine?

Tetrabenazine is a generic prescription drug used to reduce involuntary movements in patients with Huntington's disease, a progressive condition in which nerves in the brain degenerate and the person loses the ability to control their thoughts, emotions, or movements. The involuntary movements associated with Huntington's are sporadic and irregular movements that are not repetitive, but often seem to be purposeful.

Tetrabenazine works by reducing a chemical, called dopamine, that transmits signals between nerves. This reduces the ability of the brain to send signals to muscles that cause involuntary movements.

After slowly increasing the dose as directed by the doctor, Tetrabenazine is generally taken as a tablet two or three times per day. The dose will be based on reducing involuntary movements as much as possible using a dose that can be tolerated.

What are the side effects of Tetrabenazine?

Common side effects of Tetrabenazine include, but are not limited to, nausea, drowsiness, sedation, sleepiness, insomnia, tremors, slowed movement, headache, dizziness, balance problems, falls, and cold symptoms. Some serious side effects of Tetrabenazine are muscle stiffness and fast or uneven heart rate.

Other serious side effects include mental and mood changes such as anxiety, restlessness, agitation, irritability, mood changes, behavior changes, depression, severe depression, suicidal thoughts, and suicidal behavior (suicidality). Tetrabenazine has a black box warning, the most serious warning required by the FDA (US Food and Drug Administration). The warning states that Tetrabenazine can increase the risk of depression and suicidal thoughts and behavior in people with Huntington’s disease. People who take Tetrabenazine must be closely monitored for behavior changes, depression, and suicidal thoughts and behavior. Patients, caregivers, and families should understand these potential risks and report concerning behaviors immediately to the doctor. Tetrabenazine is contraindicated (should not be used) in people who are suicidal or who have depression that is untreated or inadequately treated.

Allergic reactions are rare. If you develop hives, shortness of breath, or swelling around the lips, tongue, or face, get emergency medical help right away.
